The two drivers received minor injuries and were assessed at the scene.
The accident happened as the cows were on Outram-Mosgiel Rd at about 5.55am, Sergeant Matt Lee, of Dunedin, said.
The first driver did not see the cows and struck one of them.
The second cow ran behind that car and was hit by a second car travelling in the same direction.
Both cows were severely injured, although one was able to run off the road.
The other cow was unable to move itself from the road, Sgt Lee said.
One lane of traffic was blocked until the farmer could come and attend to the cow in the road.
