Police were called to the White Horse Inn, in Union St, Milton, at 4.15pm following reports of a robbery.
The worker was getting into a vehicle when he was accosted by a male in the car park at the rear of the building and the banking taken, Sergeant Craig Dinnissen, of Balclutha, said.
The offender was spotted by a witness running from the rear of the car park along Spenser St towards Cowper St.
He was wearing a black hoodie with the hood pulled up tight around his face.
Sgt Dinnissen said an undisclosed amount of cash was taken in the robbery. He was not in a position to discuss whether a weapon was used.
The victim was uninjured but extremely shaken, he said.
A large police presence, including police dog units, could be seen in the area, while the neighbouring business, Clutha Liquorland, was taped off during the investigation.
Police were seeking assistance from the public in relation to the robbery, and anyone with information was asked to call their local police station, Balclutha police on 418-0203 or anonymously via the National Crimestoppers line 0800 555-111.
