Lock Will Tucker has come down from Canterbury and will lock the scrum with Josh Hill, forming a young second-row combination.
Hill has played just seven games for Otago, started just two matches, while Tucker has had just the four outings for Canterbury.
Hill has looked the goods at this level, but the duo will be up against some experienced and quality Auckland locks in Scott Scrafton and Highlanders second rower Jack Whetton.
Irish lock Jack Regan has been named in the reserves and will also make his Otago debut should be get on.
Charles Elton, who played his 100th game for Harbour earlier this season, will make his Otago debut at this level, playing on the blindside flank.
He was the North Otago player of the year last season, helping North Otago to win the Heartland Championship and has been a standout for his Harbour club side in recent years.
But this is a real test for Elton, who will have to stand tall against what appears to be a strong and powerful Auckland side.
Veteran Josh Hohneck will start the game at prop, his first game at this level in New Zealand since the end of 2016.
Josh Ioane comes right back into the starting side at first five-eighth while Aleki Morris-Lome is outside him. Morris-Lome had been out for a couple of months with a finger injury but impressed for Harbour in his return to the field last Saturday.
Josh Timu will start at centre, having played in that position only once previously — against Northland in 2017. He will have the big task of marking All Black Rieko Ioane.
Speedsters Vilimoni Koroi and Jona Nareki are on the wing but will have their hands full taking on new All Black Caleb Clarke and AJ Lam.
Otago winger Freedom Vahaakolo will come off the bench and will make his first-class debut should he get off the field.
He should know the opposition, having played club rugby in Auckland over the past couple of years.
Otago will be going for a hat-trick of wins over Auckland after picking up victories in 2017 and 2018. The two sides did not play each other last year.
The match is for the Lin Colling Trophy which is held by Otago.
