Centre Brett Mather and wingers Ryan Shortland and Fetu'u Vainikolo all left the field during the side's loss to Waikato last Saturday.
None of the players trained yesterday and their chances of playing on Saturday are 50-50 at best.
Mather, who injured his groin and did not come out after half-time in the match against Waikato, looks most unlikely to play.
Winger Shortland injured a hamstring and was replaced by Vainikolo, who then had to leave the field after 15 minutes with a sore knee.
Otago assistant coach Bruce Carvell said Mather was probably the most in doubt, as groin injuries did not heal easily. He was more hopeful the other two would be fit to play.
Shortland had troubles with the hamstring in the club season but has been mostly injury-free for Otago.
The former New Zealand Warrior has impressed on the wing, showing plenty of pace and not being caught out of position.
Vainikolo has struggled this year, after being a stand-out last year for the Highlanders and Northland, and is running out of games to impress selectors for next year.
Carvell said with just two games to play in the season, players could play without being fully fit, as they had the off-season to recover.
Green Island winger Jeremy Cave trained with the squad yesterday. Cave had a poor run with injuries this season, taking a couple of knocks to the head, but is a capable player with plenty of pace and size.
He was selected for a New Zealand sevens squad earlier in the season but was forced out because of a knee injury.
If all three injured players fail to recover, Otago may be forced to play midfield back Andrew Parata on the wing.
Carvell said despite the side being out of the semifinal picture, spirits were high in the squad.
