Hertz country manager Mark Righton said the new facilities, officially opened on Wednesday, included a fourfold increase in the number of parking bays and double the space for car preparation and maintenance, which would allow the company to base and prepare more vehicles in the resort.
Mr Righton said Hertz's operations in Queenstown were increasing and the facility would enable the business to be more efficient in meeting clients' needs.
The new facility is the largest car rental base in Queenstown, with parking for 130 cars and expanded cleaning and maintenance areas.
All Hertz operations across the Wakatipu Basin would be controlled from the new site, Mr Righton said.
Queenstown Airport chief executive Scott Paterson welcomed the expansion, which coincided with a 29% increase in flights to the airport this winter.
"Queenstown Airport is focused on supporting its tenants [to] manage the growth of their business and we are delighted that Hertz is opening this major service centre during our own period of growth.
"The opening of this major service centre recognises the huge potential that Queenstown offers for tourism operators such as Hertz," Mr Paterson said.
