The couple own two farms - one at Pukepito near Balclutha, and the other at Berwick.
Mr and Mrs Moore live on the Pukepito farm, dubbed Riverview due to its proximity to the Clutha River.
Mr Moore's grandfather, George Moore, bought the farm from the estate of William Pillans in 1892.
Two large floods in 1917 and 1978 had a significant effect on the farm, flooding the lower flats, Mr Moore said.
The flats were completely submerged in the 1978 flood, and stock had to be removed by helicopter and boat to prevent huge losses.
''We've had our struggles and that made it all the more worthwhile,'' Mrs Moore said.
About 1300 Romney ewes and 60 beef cows were run on the 165ha farm until the couple converted to dairy in 1996. Now, the dairy farm is now leased out to milkers and runs 350 cows.
Mrs Moore said receiving the award was on her ''bucket list''.
''I felt it really was an achievement. We're the only ones left on the original Manuka Island Estate. It just did seem an achievement.''
The Moores are also receiving a special sesquicentennial award (150 years) for their Berwick farm.
Mr Moore's great-grandparents, Donald and Anne Campbell, bought 123ha in June 1862.
Now, 58ha of the original farm remains, and is farmed by Nicholas and Nadene Moore.
Eight members of the Moore family will attend the awards, including the couple's three children - Chris, Nicholas, and Emma.
The awards are in their eighth year. The first awards night was held in 2006 and 55 families from throughout New Zealand were honoured after farming the same piece of land for 100 years or more.
Twenty-eight families will attend tomorrow night's awards function at the Simpson Park complex.
Primary Industries Minister Nathan Guy will be guest speaker, and radio farming personality Dominic George will compere the awards.
