Mr Davis, owner of Wanaka Placemakers, had his arm twisted by his Invercargill counterpart, Alistair Rance, to take up the scooter challenge, which has attracted about 50 entries.
Nicola and Graeme Vallance, of Wanaka, and Greg King will join Mr Davis on his journey, while Mark Gould will drive a support vehicle.
Last year, Placemakers franchises around the country raised about $400,000 for the foundation, Mr Davis said.
"There is some friendly rivalry to see who can raise the most money."
The scooters will leave Bluff about 8am and should arrive in Queenstown about 3pm.
